Integrated Immune Cell-Cell Communication Networks Underpinning Rheumatoid Arthritis Pathogenesis

چکیده
Rheumatoid Arthritis (RA) is a chronic, systemic auto-inflammatory disease that targets peripheral joints causing bone erosion, worsened mobility, and lower quality of life. RA complex autoimmune develops through the combined actions many cell types. In particular, monocytes, fibroblasts, T cells play critical roles within arthritic joint driving pathogenesis. These communicate with each other via ligand-receptor interactions orchestrating inflammatory responses ultimately leading to destruction. However, it remains unclear how efforts these different form an integrated network immune which lead development. Therefore, we hypothesized communication between found functions promote inflammation drive I utilized predictive assessment cell-cell networks single-cell RNA-sequencing determine differ non-autoimmune studies identified novel in fibroblasts cooperate interactions, activation, induction pro-inflammatory gene networks. Together, results identify numerous potential therapeutic for intervention disease.

Rheumatoid arthritis (RA) is a chronic inflammatory disease characterized by hyperplasia of the synovial lining cells, angiogenesis, and infiltration of mononuclear cells resulting in pannus formation, cartilage erosion and ultimately joint destruction. Synovial tissue (ST) fibroblast hyperplasia is reminiscent of tumor-like proliferation and is a major cause of cartilage destruction in the RA ...

Classically, B-cells have been considered to play a secondary role in the pathogenesis of rheumatoid artritis, restricted to the production of auto-antibodies. Nevertheless, the unexpected good clinical response that the systemic depletion of B-cells has demonstrated in a well-controlled clinical trial in patients with rheumatoid artritis has revitalized the interest in this cell type in the pa...

Rheumatoid arthritis (RA) is an autoimmune systemic disease that primarily affects joints. Etiology and the pathogenesis of RA are complex, involving many types of cells, among others macrophages, T and B cells, fibro- blasts, chondrocytes and dendritic cells. Despite well documented role of many genes and epigenetic modifications in the development and evolution of the disease, in most RA pati...
